Glycochenodeoxycholate (GCDA), a toxic component in bile salts, is involved carcinogenesis of gastrointestinal tumors. The objective this research was to study the function ERK1/2 GCDA-mediated survival and drug-resistance hepatocellular carcinoma cells (HCCs). Firstly, extracellular signal-regulated kinase 1/2 (ERK1/2) detected extensively expressed liver cancer cells, silencing by RNA interference could suppress GCDA-stimulated promote apoptosis. Furthermore, phosphorylation endogenous be potently stimulated GCDA combination with enhanced chemoresistance QGY-7703 cells. proliferation impaired PD98059, which acted as an inhibitor block ERK1/2. Mechanistically, PD98059 able nuclear aggregation p-ERK1/2, upregulate pro-survival protein Mcl-1 downregulate pro-apoptotic Bim. results indicated that disruption blocking or translocation may put forward new methods for solving problem GCDA-related treatment.
